Abstract:

In this study, the full length of turbot NPY cDNA was cloned using RT-PCR and rapid-amplification of cDNA ends (RACE). Turbot NPY cDNA is 729bp with a 5´-untranslated region (UTR), 135bp 3´-UTR and 299bp ORF. The open reading frame encodes a 99 amino acid NPY. The first 28 amino acids constitute the signal sequence followed by the 36 amino acid mature peptide. Turbot NPY share a 53.9–98.0% identity with NPY from other fish, especially, Turbot NPY share a 97–98.0% identity with NPY from other flounder (Japanese flounder, winter flounder).Using real-time PCR to analysis NPY expression in turbot. Tissue distribution studies show that NPY mRNA is expressed in brain, in particular the hypothalamic, and in peripheral tissues. During food deprivation, the NPY undergo significant change in expression (P<0.05), with NPY levels being elevated first, then decreased, and being significantly increased, maintain a high level of expression, then decreased again during food deprivation. Expression level was significantly higher with refeeding after fasting than the normal feeding, the longer of food deprivation, the higher of expression level. But the expression level of 2h after refeeding will fall again to a similar level. Our results suggest that NPY is conserved peptides that might be involved in the regulation of feeding and other physiological functions in turbot.

Molecular engineering of dendrimer nanovectors for siRNA delivery and gene silencing

Abstract: Small interfering RNA (siRNA) therapeutics hold great promise to treat a variety of diseases, as long as they can be delivered safely and effectively into cells. Dendrimers are appealing vectors for siRNA delivery by virtue of their well-defined molecular architecture and multivalent cooperativity. However, the clinical translation of RNA therapeutics mediated by dendrimer delivery is hampered by the lack of dendrimers that are of high quality to meet good manufacturing practice standard. In this context, we have developed small amphiphilic dendrimers that self-assemble into supramolecular structures, which mimic high-generation dendrimers synthesized with covalent construction, yet are easy to produce in large amount and superior quality. Indeed, the concept of supramolecular dendrimers has proved to be very promising, and has opened up a new avenue for dendrimer-mediated siRNA delivery. A series of self-assembling supramolecular dendrimers have consequently been established, some of them out-performing the currently available nonviral vectors in delivering siRNA to various cell types and , including human primary cells and stem cells. This short review presents a brief introduction to RNAi therapeutics, the obstacles to their delivery and the advantages of dendrimer delivery vectors as well as our bio-inspired structurally flexible dendrimers for siRNA delivery. We then highlight our efforts in creating self-assembling amphiphilic dendrimers to construct supramolecular dendrimer nanosystems for effective siRNA delivery as well as the related structural alterations to enhance delivery efficiency. The advent of self-assembling supramolecular dendrimer nanovectors holds great promise and heralds a new era of dendrimer-mediated delivery of RNA therapeutics in biomedical applications.

Abstract: Understanding the spatial variability of household carbon emissions is necessary for formulating sustainable and low-carbon energy policy. However, data on household carbon emissions is limited in China, the world’s largest greenhouse gases emitter. This study quantifies and maps household carbon emissions in Shanghai using a city-wide household survey. The findings reveal substantial spatial variability in household carbon emissions, especially in transport-related emissions. Low emission clusters are founded in Hongkou, Xuhui, Luwan, Jinshan, and Fengxian. High emission clusters are located in Jiading and Pudong. Overall, the spatial pattern of household carbon emissions in Shanghai is donut-shaped: lowest in the urban core, increasing in the surrounding suburban areas, and declining again in the urban fringe and rural regions. The household emissions are correlated with a number of housing and socioeconomic factors, including car ownership, type of dwelling, size of dwelling, age of dwelling, and income. The findings underscore the importance of a localized approach to low-carbon policy-making and implementation.
